I have never heard of this saying, but I am sure it relates to Andrew Duncan, who was a Scot and led in treatment of people with mental probs. Hence the "single fare", in other words going one way only! See this:-

In the Royal Edinburgh Hospital the Andrew Duncan ward was for the treatment of alcoholics and mental cases but I think it might be the alcoholic side that is referred to by you and is the most often implied when used bt Edinburgh locals.
