If you haven't got an overdraft facility, yes they will. If you haven't got one, get one - it needn't be major, just enough to act as a buffer for when incomings don't meet outgoings.
The money may or may not be taken from your account, but your bank will charge you either way - either for an unauthorised overdraft or for declining to pay the direct debit.

Your debit card transaction will go through and your bank will charge you for unauthorised borrowing. However, at the time the transaction was processed you must have had sufficient funds in your account otherwise the transaction would have been declined. Through an epos system a debit card transaction is instantaneous. There is no "clearing " period as with a cheque.

how do you not know how much you have in your account??? Wouldnt it have been wise to check before purchasing

hold on a minute...

you paid by debit card. It will probably just get declined if it is processed manually later (such as a small firm) and the company may ring/email to get alternative payment details. If the transaction was approved when you ordered online, then yes it will push you o/d and you may or may not be charged, depending on how much you have gone over your limit. Some banks offer a small buffer (10 or so) within which you will not be charged.
