Can I microwave a homemade lasagne from frozen or do I need to thaw it first?

I don't see why not so long as it is hot all the way through - there are loads of shop bought frozen ones that you cook straight from the freezer

I always microwave from frozen. If it's just a single sized portion, seven minutes should do it.
