I am going to put tiles above the upstand on the kitchen worktop.
Do I put the first row of tiles directly on the upstand top or put a spacer in?
Thanks in advance.

If you're just continuing what's there already, Needy, then definitely a spacer.

Make sure you're confident of the existing tiles being straight. If they're not, you'll need to start from the next row up... leaving a gap to be filled in afterwards. I'll explain it better if you need to do that.

Always the first Builder!
At present there are no tiles at all. Just the upstand. So do I put a spacer in between the upstand and the first row of tiles or the tile straight on top of the upstand?
Hiya Needy.
Sorry, I thought your upstand was existing tiles. :o(((

Assuming the upstand is nice and straight, then set them directly onto it, without the spacers.

If you do that, it's a good idea to run a thin silicone bead along afterwards to cover the joint.

If you don't want silicone, then set them on spacers so the joint can be grouted with the rest.
