We have to remove a massive tree root which is raising the level between the road and the garage. If this kills the tree, so be it. But we would rather it didn't, so is it best to coat the wound and, if so, what with?

Getting rid of one tree root will not necessarily kill the tree. I don't know what you could coat the wound with sorry.
It might not kill the tree, but it might make the tree fall over in a high wind.
A "massive" root is probably quite an important part of the tree's support. Can you not get professional ( tree-surgeon) advice ?
