How would a garage conversion affect the value of our 3 bed semi. We do have off road parking.

Why not run the scenario past a couple of local estate agents - make sure you start the conversation by telling them you are thinking of selling, or they won't be as keen to give you free advice - and you should get an idea of the market in your area, and the viability of your proposal.
Losing a garage can often be detrimental to value regardless of what you convert it into. It may sound daft these days, when most of us don't use garages to keep our cars in, but they are still a big selling point for houses. Also, garage conversions can be difficult to do right from an aesthetic point of view. This could cause a problem too. On the flip side, if doing this will make a significant difference to your downstairs area, then it may be worthwhile. Talk to estate agents; they know what sells.
We have bought a house recently where the garage had already been converted into a study/office with just a store room type of space left big enough for my fridge/freezer and gardening stuff. We have only one car which we park on the driveway but it can be off putting to people with more cars. Having to park on the road can put people off because of other types of vehicles passing by or the need of a permit. We love the study - in fact thats where I'm sitting typing my reply now!!!
