Can I send a letter first class using one first class stamp and one second? I.e. if it is a large letter which will cost 52p to send first class, can I use one first and one second to meet the price?

Yes - but you will be overpaying. 36+27 is 63.

In general 1st and 2nd class stamps can be used at their current 'face' value to make up any amount for either 1st or 2nd class mail, and most of the time will be handled correctly.
