It depends on where the musical was on and what the show is. If you mean a high-profile West End show, opening nights are often for Press only, and any other tickets there are will sell like hot cakes, but you can always try getting them from Ticketmaster or through the theatre. Generally, though, buying tickets for opening nights is no different than buying them for any other night. I work in one of the main theatres in Birmingham, and, while opening nights are always very popular, if you book enough in advance you should be able to get tickets.
New shows often have websites in advance, so you could go to the site and email the person running it- who is usually an obsessive fan! They will know everything and everybody, and be a good "human" contact!
