If you are able to plug a laptop into the mains when using it, is that the best thing to do?

Alternatively should you run it on batteries, and just recharge it when it needs it?

You will bugger it up if you keep it plugged in all the time. Run it on batteries and charge when necessary.

I don't mean leave it plugged in all the time, just plug it in while using it.

Would that still harm it?
That's what I thought you meant - sorry. No don't have it plugged in all the time you're using it. You need to let the battery run down on a regular basis. When I see mine getting a bit low I plug it in for a while.

Battery will last longer if you charge it when necessary. Applies to most appliances that run off rechargeable batteries.
