quite easy, get the train in madrid to paris, then the eurostar from paris to london, waterloo
it cannot be done in one day - the earliest you can get to paris from madrid is 22h00 at night, (dep 06h25 via barcelona & montpellier), after the last eurostar has left for london. see http://www.seat61.com/Spain.htm for ways of travelling between spain & london.

As Mushroom25 states, it can't be done in a day.

Here are some specimen times, based on travel this coming Monday:

Depart Madrid Chamartin at 1000 (There are no earlier trains, on Monday, which provide useful connections. Mushroom25 must have checked a different day).
Arrive Hendaye at 1656. (There is no direct service from Madrid to Paris).

Depart Hendaye at 1722.
Arrive at Paris Montparnasse at 2315.

Metro or taxi to Paris Nord. (I'm not actually sure whether you can stay in Gare du Nord station overnight. I know that Gare de l'Est shuts for a few hours).

Depart Paris Nord 0622
Arrive Waterloo International 0758.

Those times are taken from the Deutsche Bahn website, which allows you to plan journeys anywhere in Europe:
