Start - control panel - performance and maintenance.
Free up space on hard drive
Then
Rearrange items on hard drive.
The second is the actual defrag, but it is better to do the first before hand.
Be prepared for it to take an age, but if you do it every three months or so, it won't be so bad in future.

Click Start >>Programs>>Accessories>>System Tools>>'click' Disc Degragmenter
From the window that pops up select your HDD normally listed as (C) as default, then click Analyse and Defragment.
If this is a first Defrag or one that has not been done for some time it can take a long time! Sometimes it's best to set it going overnight after you've finished with the pc. Close down any open programs because it slows down the process.
There are other reasons for a slow pc, but thats another question! :)
