Is it true that folding a cheque makes it take longer to go through the system and clear, or is this one of those "old wives tales"?

Sort of.

If a cheque is folded too much it may not be automatically read by the machine, gets spat out and is processed manually.
We used to tear a little piece from the cheque number, that would mean the cheque had to manually processed ( for the same reason as Ethel's answer )
It shouldn't take any longer, just a little more work for the people processing it.
