If you go into Google maps it is easy to work it out yourself.
Find Manchester and RIGHT click then select "Directions from here" then find say "Warwick" and again right click and select "Directions to here".
That gives you the distance in miles and the driving time.
Do the same from Southampton and play around with the various "routes" to find a central one.

According to Google maps it is 234 miles from centre of Manchester to centre of Southampton. 4 hours driving time.
This is down the M6, round Birmingham, then down M40 then A34 to Southampton.
So you want something about 2 hours driving time from each place.
As has been said, Warwick fits the bill (it is on the M40 as well so easy to get to).
If you want something larger nearby then maybe Stratford upon Avon (lovely town with lots to do), or Coventry (not such a lovely town).
