I have been asked to set a quiz, there has to be 40 General Knowledge questions, I have never done one before so out of 40 how many need to be getable and how many to sort the men from the boys so to speak.

I also set quizzes on a fairly regular basis and agree that it is important that the difficulty level of questions varies, to ensure that the weaker teams are not disillusioned whilst also providing a reasonable test for the more able quizzers.
There is no hard and fast rule, but I would normally aim to set approx 25% easy, 50% middle of road and 25% more testing questions.
You may also wish to break the quiz into sub sections with perhaps 5 questions on specific categories.
