have just downloaded this via the link given on Piggynose's thread. On the setting window everything is ticked except 'Terminate OE during threat removal' - should I do this and why?
And am informed 'File assassin can help delete locked files' - do I run this tool?

File Assasain is a tool for deleting files which are locked. Malwarebytes will do that anyway for any files related to malware, so I wouldn't have thought you needed to run it separately.
Terminating IE would be sensible because some malware has been known to install what used to be called "helper apps" in IE. Still if IE isn't your default browser that may explain why the option isn't ticked.

Remember to update malwarebytes before running. There is a tab to do this.
