Check that the line in has not been muted. (by default it normally is) If you have a little speaker on your task bar simply double click it and the windows mixer should appear. Uncheck the mute box and all should be well.
If you have not got a speaker on your taskbar go to the control panel, double click sounds and audio and select the advanced button. The mixer will appear then.

As suggested by someone else, it could be a faulty soundcard, but before replacing it there's 2 things that might be worth trying.
1) Update the drivers for your card. It may be that they are out of date or corrupted. If you Google "Your soundcard brand/model" and "driver" you should be able to see if there is anything newer.
2) Go to Start>All Programs>Accessories>Entertainment and open "Sound Recorder". Go to Edit>Audio Properties and check that your soundcard is the default recording device. Also click the Volume button and double check that the "select" box for "Line In" is checked (you'll probably also have "CD Player" and "Microphone" options). Press Play on your tape deck and try to record its output. You have a max of 60 seconds and the controls should be self explanatory. Try playing back your recording. If you have recorded something then it is likely to be a problem with your other program or its set up. If you're still not recording anything, assuming all the hardware is connected properly and there are no "mute" buttons pressed in the software, I would look to a hardware problem, either with the connecting leads or your soundcard.
Do you know anyone else with a computer? If you try idea (2) on their machine, but with your tape deck and lead and can record, then you can rule out the fault being external to your PC.
